Ultrasound-targeted nanoparticle delivery across the blood-brain barrier

چکیده

Background/introduction The delivery of systemically administered drugs and genes to the CNS is hindered by both the blood-brain barrier (BBB), which limits transport from the bloodstream to the brain to only a few privileged molecules, and the nanoporous electrostatically charged tissue space, denoted here as the “brain tissue barrier” (BTB). Our group engineers targeted drug and gene delivery approaches, capable of overcoming both of these physical barriers, for the treatment of brain tumors and neurodegenerative diseases. We focus on nanoparticle (NP) delivery systems, as they offer the potential for enhanced transfection efficiencies and controlled-drug release.

The effect of nanoparticle size on the probability to cross the blood-brain barrier: an in-vitro endothelial cell model

BACKGROUND During the last decade nanoparticles have gained attention as promising drug delivery agents that can transport through the blood brain barrier. Recently, several studies have demonstrated that specifically targeted nanoparticles which carry a large payload of therapeutic agents can effectively enhance therapeutic agent delivery to the brain. However, it is difficult to draw definite...

Lipid microbubbles as a vehicle for targeted drug delivery using focused ultrasound-induced blood-brain barrier opening.

Focused ultrasound in conjunction with lipid microbubbles has fully demonstrated its ability to induce non-invasive, transient, and reversible blood-brain barrier opening. This study was aimed at testing the feasibility of our lipid-coated microbubbles as a vector for targeted drug delivery in the treatment of central nervous system diseases. These microbubbles were labeled with the fluorophore...

Improved anti-tumor effect of liposomal doxorubicin after targeted blood-brain barrier disruption by MRI-guided focused ultrasound in rat glioma.

The blood-brain barrier (BBB) inhibits the entry of the majority of chemotherapeutic agents into the brain. Previous studies have illustrated the feasibility of drug delivery across the BBB using focused ultrasound (FUS) and microbubbles. Here, we investigated the effect of FUS-enhanced delivery of doxorubicin on survival in rats with and 9L gliosarcoma cells inoculated in the brain. Each rat r...

A New Brain Drug Delivery Strategy: Focused Ultrasound-Enhanced Intranasal Drug Delivery

Central nervous system (CNS) diseases are difficult to treat because of the blood-brain barrier (BBB), which prevents most drugs from entering into the brain. Intranasal (i.n.) administration is a promising approach for drug delivery to the brain, bypassing the BBB; however, its application has been restricted to particularly potent substances and it does not offer localized delivery to specifi...
